MOTS-c and DSIP: two under-discussed ones, what do we actually know?

These two rarely get their own threads so I am bundling them, with links to both Library pages. Different mechanisms, but both suffer from the same problem: interesting biology, very little human data.

MOTS-c is a mitochondrial-derived peptide with genuinely fascinating basic-science work around metabolic regulation and exercise mimetic effects, mostly in rodents. DSIP (delta sleep-inducing peptide) has been studied since the 70s in the context of sleep but the human findings are old, inconsistent, and honestly kind of murky about whether it reliably does anything.

For anyone who has read deeper than I have: is there any recent human work on either that moves the needle, or are we still firmly in 'promising mechanism, no clinical answer' territory?

Still firmly in that territory for both, as far as I can find. MOTS-c is legitimately exciting as endogenous biology and there is real academic interest in mitochondrial-derived peptides, but the translation to a supplement people inject is a huge unproven leap. The exercise-mimetic framing in particular is doing a lot of work that the human data cannot back yet.

DSIP is almost a cautionary tale: half a century of scattered research and still no clear consensus that exogenous DSIP reliably improves sleep in humans. When something has been studied that long without a solid answer, that itself is information.

Glad someone made this thread. I will only add, as personal experience and not advice, that the DSIP sleep effect I chased was underwhelming and inconsistent enough that I could not distinguish it from placebo or from just fixing my sleep timing. One anecdote, low confidence, but it lines up with how muddy the literature is.
