MOTS-c keeps getting called an 'exercise mimetic,' and I want to pressure-test how much that label is earned versus a catchy summary.

The underlying biology is genuinely neat: it's a mitochondrial-derived peptide encoded in the 12S rRNA region, it translocates to the nucleus under metabolic stress, and it influences AMPK signaling and metabolic homeostasis. In mouse work it's been associated with improved insulin sensitivity and exercise capacity, which is where the 'mimetic' branding comes from.

My hesitation: most of the striking functional data is rodent, and 'affects the same pathway exercise does' is not the same as 'reproduces the benefits of exercise in humans.' Has anyone seen human data beyond observational/associative work on circulating MOTS-c levels? I'd like to calibrate how much weight the label deserves.

I think 'exercise mimetic' is best read as a mechanistic hypothesis with good mouse support, not an established human phenotype. The AMPK convergence is real, and declining MOTS-c with age is an interesting association, but association isn't the same as the peptide driving the outcomes.

The thing I'd flag for newcomers: exercise hits dozens of pathways at once. Even a peptide that genuinely nudges one node isn't going to be a substitute, and framing it that way oversells what any single molecule does.

Agree the human side is thin. I've read the aging-association papers and they're suggestive but cross-sectional. Easy to build a nice narrative on top of them that the data can't actually hold up.

Sharing purely as personal reading, not a recommendation for anyone: I've stayed on the sidelines of this one precisely because the human evidence hasn't matured. The mouse metabolic results are exciting but I want to see controlled human work before I'd even form an opinion, and anyone weighing anything health-related should be talking to a clinician, not a forum.
