Is Humanin the most underrated of the mitochondrial-derived peptides?

MOTS-c and SS-31 soak up most of the mitochondrial-peptide attention here, but Humanin was the first mitochondrial-derived peptide characterized and it has a surprisingly deep literature that rarely comes up.

What draws me to it: cytoprotective and anti-apoptotic signaling, associations with cardiometabolic and neuroprotective outcomes in various models, and some human cohort data suggesting circulating Humanin declines with age and correlates with things like offspring longevity in certain studies. That combination of a plausible mechanism plus actual human association data is rarer than you'd think in this space.

My honest caveat, same as always: associations and model systems are not clinical proof, and I don't want to overstate it. But I'm curious why it gets so little airtime relative to newer peptides. Is it just that it's harder to romanticize than a shiny 'exercise mimetic'? Would love others' takes on where the evidence is strongest and weakest.

I think it's partly narrative and partly timing. Humanin came out of Alzheimer's research rather than the longevity-branding wave, so it never got the same marketing gloss. The science is arguably more mature than some of the trendier peptides precisely because it grew up in a disease-mechanism context.

Strongest evidence in my view is the mechanistic cytoprotection work and the age-decline human associations. Weakest is anything resembling an interventional human longevity trial, which basically doesn't exist. So same shape as the rest of the field: good biology, no outcome trials.

Really glad you started this. I'd add that the small-humanin-like-peptides (SHLPs) coming out of the same alt-ORF region are an interesting adjacent rabbit hole and similarly under-discussed.

As a newcomer it's clarifying to see the pattern repeat across every compound in this category: elegant mechanism, some human association data at best, and a clear 'talk to a professional, this isn't settled' asterisk. Almost feels like that should be the category's tagline.
